The findings below come from in-vitro and animal studies and, where noted, clinical trials of approved pharmaceutical products. Semaglutide is the active ingredient in FDA-approved prescription medicines (Ozempic®, Wegovy®, Rybelsus®). The research-grade reference material sold here is not a drug product, is not manufactured under pharmaceutical cGMP, and must not be used as a substitute for any approved medicine, and nothing on this page is medical advice or a therapeutic claim.
GLP-1 Receptor Agonism
Semaglutide shares roughly 94% sequence homology with native human GLP-1 and is a full agonist of the GLP-1 receptor, driving cAMP signalling, glucose-dependent insulin secretion and glucagon suppression in islet and cell-line models.
Structural Engineering for Half-Life
Aib at position 8 confers resistance to DPP-4 cleavage, and the C18 fatty diacid side chain promotes reversible albumin binding, reducing renal clearance. The discovery program reported an elimination half-life of roughly one week in humans.
Central Appetite Pathways
Rodent studies using labelled semaglutide show access to circumventricular and hypothalamic regions, including the arcuate nucleus, with downstream effects on neural pathways associated with food intake.
Metabolic Signalling Studies
In pancreatic islet and β-cell models, GLP-1 receptor activation by semaglutide raises intracellular cAMP and potentiates insulin secretion only at elevated glucose concentrations, a glucose dependence central to incretin research. Diet-induced obese rodent models are widely used to study its effects on food intake and body-weight regulation.
Oral Absorption Research
Semaglutide is also studied in oral peptide-delivery research: co-formulation with the absorption enhancer SNAC was shown to promote local, pH-buffered absorption across the gastric epithelium.
Clinical Context
Unlike most compounds in our catalog, semaglutide has extensive randomized clinical trial data, including the STEP program. That literature concerns the approved pharmaceutical products, not research-grade material.
