What Is Mazdutide?
Mazdutide (IBI362, also designated LY3305677) is a lipidated oxyntomodulin analogue (4563.06 g/mol) that acts as a dual agonist at the GLP-1 and glucagon receptors. It was developed by Innovent Biologics from a molecule originally licensed from Eli Lilly, and it is studied as a tool for probing combined GLP-1 and glucagon pathway activation.
Its receptor profile is what distinguishes it in the incretin field. Tirzepatide pairs GLP-1 agonism with GIP; mazdutide pairs it with glucagon, a receptor associated in the literature with increased energy expenditure rather than insulin secretion. An Aib substitution at position 2 blocks DPP-4 cleavage, and a fatty-acyl chain on Lys20 — attached through two AEEA spacers and a γGlu linker — provides albumin binding and once-weekly kinetics.
Supplied as 10 mg of white lyophilized powder at ≥99% HPLC purity with a batch-specific third-party COA. The first production batch is in final QC; the form below will bring you the full analytical package as soon as it is released.

GLP-1 / Glucagon Dual Agonism
Mazdutide activates the GLP-1 receptor and the glucagon receptor (GCGR). This is the key distinction from tirzepatide, which pairs GLP-1 with GIP instead. In vitro work describes potent binding at both human and mouse GLP-1 and glucagon receptors. Mazdutide is not a GLP-2 agonist.
Oxyntomodulin Scaffold
Unlike semaglutide, which is built on GLP-1(7–37), mazdutide derives from oxyntomodulin — an endogenous proglucagon product that natively engages both the GLP-1 and glucagon receptors. The molecule is effectively a stabilized, lipidated version of that dual-acting hormone.
Preclinical and Early Clinical Reports
A phase 1b multiple-ascending-dose trial in Chinese adults with overweight or obesity reported body-weight reduction of up to about 6.4% over 12 weeks at doses up to 6 mg. A separate phase 1b type 2 diabetes cohort reported reductions in HbA1c and fasting plasma glucose from baseline to week 12.
Tolerability Signal
Gastrointestinal events dominated the adverse-event profile in early trials, with diarrhoea reported in roughly 29% of IBI362 recipients against 33% for dulaglutide and 0% for placebo. The absence of a GIP arm has been discussed as a possible contributor to the tolerability differences between dual agonists.
Limitations
The phase 3 GLORY-1 figures circulating online come largely from secondary sources and should be checked against the primary publication. Regulatory status reporting also conflicts between sources, so treat any single claim with caution.

Reconstitution Procedure
Allow the vial to reach room temperature. Introduce sterile or bacteriostatic water slowly down the inner glass wall — do not inject directly onto the powder cake. Swirl gently until fully dissolved; do not shake or vortex, as this can promote aggregation.
